This prospective study evaluated the feasibility of biotin labeling for tracking caprine red blood cells (RBCs) and assessed posttransfusion survival of fresh versus stored allogeneic whole blood in healthy goats. Fifteen healthy goats (13–15 months old) were assigned to three groups of five, each serving as both donor and recipient. Group 1 received fresh biotin-labeled whole blood (within 6 hours of collection), Group 2 received blood stored at 4°C for 14 days, and Group 3 received blood stored at 4°C for 28 days. Flow cytometry successfully distinguished biotin-labeled RBCs from unlabeled cells, confirming the viability of this labeling technique in goats. Serial blood sampling over 28 days post-transfusion allowed nonlinear regression modeling to determine RBC half-lives. Results demonstrated half-lives of 4.6, 4.3, and 2.5 days for Groups 1, 2, and 3, respectively. Using Akaike information criterion (AIC) comparisons, Groups 1 and 2 showed no significant difference in RBC survival, while Group 3 demonstrated a significantly reduced half-life compared to both. These findings indicate that storage up to 14 days does not significantly impair posttransfusion RBC survival in goats, but 28-day stored blood is associated with markedly reduced RBC longevity. This study establishes important baseline data for caprine transfusion medicine and suggests that clinicians should consider storage duration when administering blood transfusions in goats, particularly avoiding blood stored for 28 or more days when optimal RBC survival is clinically important.
