This prospective in vitro study evaluated canine platelet concentrate (PC) units stored at 4°C for 14 days without agitation. Six PC units were analyzed on Days 0, 5, 7, 9, 12, and 14 using platelet counts, indices, metabolic markers, aggregometry, flow cytometry, and bacterial cultures. Platelet counts remained stable (>500,000/µL), and all pH values stayed >7.1. Lactate increased significantly from 2.1 to 9.6 mmol/L by Day 14. While platelets remained viable, platelet function demonstrated agonist-dependent deterioration: ADP response was minimal, thrombin-induced aggregation remained robust until Day 12, collagen showed poor response throughout, and convulxin induced significant activation. Alpha-granule secretion increased on Days 12-14 but decreased after Day 7 with thrombin stimulation. Flow cytometry showed variable P-selectin and Annexin V expression patterns. One sample developed bacterial growth by Day 14. The findings indicate that cold storage at 4°C can maintain platelet viability and basic hematologic parameters while demonstrating predictable functional changes, supporting further investigation as a potential strategy to extend canine PC shelf life beyond conventional room temperature storage.
