Summary

This study evaluated the safety profile of izenivetmab, a caninized monoclonal antibody targeting nerve growth factor (NGF), for treatment of canine osteoarthritis pain. Three laboratory studies were conducted in adult Beagle dogs assessing general health, clinical pathology, toxicokinetics, and immunogenicity. Study 1 included comprehensive evaluations (electrocardiography, neurologic, ophthalmic, radiography, and histopathology of joint, nervous, and immune tissues). Study 2 evaluated higher dose multiples administered monthly. Study 3 assessed functional immune response. Izenivetmab demonstrated excellent tolerability at doses up to 120X the intended clinical dose (0.05-0.1 mg/kg every three months). Treatment-emergent anti-drug antibodies occurred in 4.2-8.3% of treated dogs without safety consequences. No adverse effects were detected in cardiac, neurologic, ophthalmic, or immunologic parameters. These preclinical safety data support the use of izenivetmab as a novel analgesic therapy for canine osteoarthritis, targeting an important pain mediator pathway.
