This randomized controlled study evaluated probiotic ear drops containing Lactiplantibacillus plantarum YUN-V2.0 and Lacticaseibacillus rhamnosus YUN-S1.0 in 15 healthy dogs (5 groups, n=3 each) for treating otitis externa. Ear swabs collected at 24, 48, 72, 96 hours, and 7 days post-application demonstrated significant Lactobacillaceae engraftment, increasing from baseline 1.34 × 10 cfu/mL to 2.3 × 10 cfu/mL at 24 hours and maintaining 1.6 × 10 cfu/mL at 7 days. In vitro antimicrobial testing against clinical isolates of Pseudomonas aeruginosa, Staphylococcus pseudintermedius, and Malassezia pachydermatis revealed superior or equivalent growth inhibition compared to conventional antimicrobials (florfenicol, gentamicin, marbofloxacin, miconazole, neomycin, terbinafine). Lactobacilli showed better inhibition of P. aeruginosa versus gentamicin and marbofloxacin, and superior activity against S. pseudintermedius compared to gentamicin, neomycin, and marbofloxacin. Results support probiotic ear drops as a potential therapeutic option for canine otitis externa management through sustained colonization and pathogen inhibition.
