Ten commercially prepared canine pRBC units were coincubated with normal saline solution, dextrose, Normosol-R, fentanyl, propofol, norepinephrine, dopamine, or same-donor fresh frozen plasma as control. Two storage ages (<7 and 28-35 days) and two incubation times (3 and 30 minutes) were examined. Assessment included complete blood counts, light microscopy, methemoglobin assays, and osmotic fragility testing. Results demonstrated that normal saline caused significant mean cell volume increases under all conditions and mild RBC swelling. In older blood (>28 days), dextrose, norepinephrine, dopamine, and fentanyl also increased MCV. Most additives showed minimal deleterious effects on fresh RBCs during short coincubation periods, but older blood demonstrated mild swelling and increased osmotic fragility with most additives. Propofol's lipid emulsion created analytical interference, preventing accurate assessment of its true effects. The study highlights the need for further investigation regarding safe coadministration practices of RBCs with crystalloids and medications.
