This study demonstrates that BLOS1 overexpression in goats enhances innate immune responses against Brucella infection through multiple mechanisms. Macrophages from transgenic goats overexpressing BLOS1 showed enhanced activation of NF-κB and TLR4 signaling pathways upon Brucella LPS stimulation, resulting in increased TNF-α secretion. Critically, BLOS1 overexpression significantly upregulated autophagy-related genes (LAMP2 and BECN1), improving autophagic flux and bacterial clearance capacity. Beyond cellular immunity, BLOS1 overexpression remodeled the intestinal microbiota composition, enriching pathways associated with membrane transport and cellular motility while reducing abundance of virulence factors and opportunistic pathogens. These microbiota changes contributed to improved intestinal immune homeostasis.
