This prospective study evaluated whether oral administration of pentobarbital sodium at three times the labeled intravenous dose could induce unconsciousness or euthanasia in dogs scheduled for euthanasia in a shelter setting. Twelve dogs exhibiting human and/or animal aggression were enrolled between June 2024 and October 2025, with no restrictions on breed, sex, or age. Pentobarbital sodium solution was placed into size 00 gelatin capsules (1-mL capacity) and concealed in high-reward foods. Nine of 12 dogs consumed the food, while 3 refused. Of the 9 dogs that ingested the drug, 5 achieved unconsciousness at a dose of ≥255 mg/kg within a mean time of 25 minutes. Respiratory rates remained slow but consistent throughout the 120-minute observation period. Critically, none of the 12 dogs achieved death solely from oral pentobarbital administration, and all required a secondary euthanasia method. The findings indicate that oral pentobarbital is unreliable as a standalone euthanasia agent in dogs. However, at doses of ≥255 mg/kg, it demonstrated utility as a pre-euthanasia anesthetic in behaviorally challenging dogs where safe handling is compromised. This technique may offer a practical, lower-stress option for veterinary and shelter personnel managing aggressive dogs prior to euthanasia, though acceptance of the food vehicle remains a limiting factor and an alternative euthanasia method must always be available.
