This prospective experimental study investigated the minimum contact time required for topical difluprednate lotion to produce detectable glucocorticoid (GC) effects in dogs, addressing a clinically relevant gap in veterinary dermatology. Six male Beagle dogs were enrolled, and suppression of the histamine-induced vascular reaction (HVR) was used as a surrogate marker of topical GC activity, based on the established vasoconstrictor assay principle. Difluprednate lotion was applied to 2 cm circular areas on the lateral trunk at 5, 10, and 30 minutes prior to intradermal histamine injection. Intravenous fluorescein was administered to visualize vascular leakage under UV illumination, and HVR area was quantified using ImageJ software. Results demonstrated statistically significant suppression of HVR at all three time points (5, 10, and 30 minutes) compared to untreated control sites (p < 0.01). No significant differences in HVR suppression were observed among the three treated time points, suggesting that even 5 minutes of contact time was sufficient to produce measurable anti-inflammatory activity. These findings indicate that difluprednate lotion achieves detectable dermal GC effects rapidly after application. Clinically, this information can guide veterinarians in providing more precise contact-time instructions to clients, potentially improving treatment adherence, minimizing premature removal, and optimizing both efficacy and safety of topical GC therapy in dogs with inflammatory skin disease.
