This study evaluated the prophylactic efficacy of a canine parvovirus monoclonal antibody (CPMA) against experimental CPV-2b challenge in 7- to 8-week-old Beagle pups. Twenty pups received a single subcutaneous dose of CPMA (0.1 mL/kg) on study day 0, while 5 control pups received PBS. All pups were challenged intranasally with virulent CPV-2b one day post-treatment. Clinical monitoring over 14 days included rectal temperature, vomiting, fecal consistency, lymphocyte counts, and fecal CPV-2 shedding. Disease was defined by meeting at least 3 of 4 criteria: fever ≥39.7°C, lymphopenia ≤50% of baseline, diarrhea, and fecal virus detection. All 5 control pups met the full CPV-2 disease case definition and developed parvovirosis. In contrast, none of the 19 prophylactically treated CPMA pups met more than 1 disease criterion. One additional pup exposed to CPV-2 via environmental contamination and treated therapeutically also did not develop clinical disease signs. The results demonstrate that a single prophylactic dose of CPMA provided complete protection against experimental CPV-2 disease. These findings support a label claim for prophylactic use of CPMA, particularly in high-risk scenarios such as outbreak situations or in naïve pups with uncertain vaccination history. The passive immunity conferred by CPMA offers a practical tool when conventional vaccination timelines are insufficient or maternal antibody interference is a concern.
